=== Structure-activity experiments === When the 12-amino acid sequence of 'peptide 3'/CCL2 is aligned with the sequences of the other chemokines CCL3, CXCL8 and CXCL12 5 amino acids are conserved, Cys3, Asp5, Pro6, Trp10 and Gln12. In addition Val11 is also present in CCL3 and CXCL8. The corresponding 11th amino acid in CXCL12 is Ile. Ala4 in CCL2 is also present in CCL3 but the corresponding residue is Leu in CXCL8 and Ile in CXCL12. Inclusion of Leu at position 4 and Ile at position 11 (NH2-EICLDPKQKWIQ-OH) increased the inhibition potency of the peptide to 2-4μM. Removal of the first two residues from 'Peptide 3' (NH2-CADPKQKWVQ-OH) does not lower its BSCI potency. Peptide 3-Leu4, Ile11, the most potent BSCI peptide was chosen for further development. The first two amino acids were removed, and then according to the method of Jameson et al., the amino acid sequence was reversed and the amino acids replaced with D-amino acids. This combined transformation changes the orientation of the amides in the backbone but does not change the position of the amino acid sidechains. The peptide was then cyclised by adding a cysteine to the other end (HO-CLDPKQKWIQC-NH2), and oxidisation to form a disulfide bond to give NR58,3-14-3.

=== S phase (interphase) extract === When CaCl₂ is added to an M-phase Xenopus egg extract at a concentration sufficient to override residual EGTA (typically several hundred micromolar), it triggers rapid inactivation of maturation promoting factor (MPF). This occurs through cyclin B degradation by the proteasome and induces cell cycle progression from metaphase to anaphase, and eventually into S phase. The resulting extract is referred to as an S-phase extract or interphase extract. The relatively slow metabolism of acetonitrile to hydrogen cyanide allows more of the cyanide produced to be detoxified within the body to thiocyanate (the rhodanese pathway). == Properties == Flavin adenine dinucleotide consists of two portions: the adenine nucleotide (adenosine monophosphate) and the flavin mononucleotide (FMN) bridged together through their phosphate groups. Adenine is bound to a cyclic ribose at the 1' carbon, while phosphate is bound to the ribose at the 5' carbon to form the adenine nucleotide. Riboflavin is formed by a carbon-nitrogen (C-N) bond between the isoalloxazine and the ribitol. The phosphate group is then bound to the terminal ribose carbon, forming a FMN. Because the bond between the isoalloxazine and the ribitol is not considered to be a glycosidic bond, the flavin mononucleotide is not truly a nucleotide. This makes the dinucleotide name misleading; however, the flavin mononucleotide group is still very close to a nucleotide in its structure and chemical properties.
